炎癥性腸病(IBD)是一種難以根治的慢性腸道炎癥性疾病,并且有潛在的癌變風(fēng)險(xiǎn)。目前基于小分子藥物、抗生素和抗體的臨床治療方法并不完全有效,長(zhǎng)期使用甚至可能導(dǎo)致嚴(yán)重的并發(fā)癥、抗生素耐藥性和免疫反應(yīng)。IBD難以治療部分原因是治療缺乏特異性,大多數(shù)治療方式依賴于血液循環(huán),很少真正針對(duì)病變部位,因此,可以定位到炎癥部位的創(chuàng)新靶向治療策略非常重要。有炎癥的結(jié)腸的特點(diǎn)之一是富含帶正電的蛋白質(zhì)及活性氧(ROS)水平升高,通過帶負(fù)電荷的藥物和藥物載體來靶向這些帶正電的蛋白質(zhì),并清除ROS來減輕炎癥反應(yīng)的治療策略可能是一種非常有效的治療方法。


納米材料在生物成像、癌癥早期診斷和藥物運(yùn)載等領(lǐng)域的應(yīng)用越來越廣泛,為醫(yī)學(xué)診斷和治療提供了有力的物質(zhì)基礎(chǔ)和技術(shù)支持,如何安全、有效、方便地使用納米材料是納米醫(yī)學(xué)的最大難題之一。納米二氧化鈰是一種具有超氧化物歧化酶和過氧化氫酶樣活性且可以清除ROS的多酶模擬物,催化抗氧化活性使納米二氧化鈰能夠通過消除ROS而有效地治療各種疾病,包括炎癥性疾病。與CeO2納米顆粒不同,蒙脫土(MMT)不能通過胃腸道屏障,也不會(huì)干擾正常的結(jié)腸功能。近期南京大學(xué)魏輝教授課題組在“Advanced Functional Materials”上發(fā)表了題為“An Orally Administered CeO2@ Montmorillonite Nanozyme Targets Inflammation for Inflammatory Bowel Disease Therapy”的文章,結(jié)合CeO2納米顆粒和MMT獨(dú)特的理化性質(zhì),設(shè)計(jì)了一種可以口服給藥的系統(tǒng),用于IBD的治療且獲得了顯著的效果。與基于酶的療法相比,納米鈰具有穩(wěn)定性好、多酶活性和免疫原性低等優(yōu)勢(shì)。CeO2@MMT通過顯著下調(diào)促炎細(xì)胞因子水平和同時(shí)上調(diào)抗炎細(xì)胞因子水平來調(diào)節(jié)病變組織的局部免疫環(huán)境。


本研究使用了欣博盛小鼠IL-1β 和 IL-10 ELISA試劑盒,評(píng)價(jià)了小鼠結(jié)腸組織勻漿中促炎和抗炎細(xì)胞因子的水平。
