心肌梗死(MI)是世界范圍內(nèi)導(dǎo)致死亡和殘疾的主要原因之一,炎癥反應(yīng)在MI修復(fù)中起著關(guān)鍵作用。心肌梗死后,大量中性粒細(xì)胞、單核細(xì)胞和巨噬細(xì)胞在受損心肌募集,以誘導(dǎo)炎癥反應(yīng)。這些炎癥細(xì)胞的持續(xù)浸潤阻礙組織修復(fù),進(jìn)而導(dǎo)致不良重塑和心功能受損。內(nèi)源性中性粒細(xì)胞凋亡的精確調(diào)節(jié)和誘導(dǎo)可能有利于促進(jìn)再生。然而,中性粒細(xì)胞固有的存活/凋亡平衡是由復(fù)雜的信號(hào)因子網(wǎng)絡(luò)監(jiān)測的,誘導(dǎo)中性粒細(xì)胞凋亡的治療可能會(huì)干擾固有免疫反應(yīng)。而且,在原位精確地實(shí)現(xiàn)中性粒細(xì)胞凋亡的誘導(dǎo)是很困難的。因此,以凋亡中性粒細(xì)胞來克服復(fù)雜炎癥微環(huán)境的替代療法也許可以有效治療心肌梗死。
近期,第四軍醫(yī)大學(xué)金巖、劉世宇教授和西安交通大學(xué)陳鑫教授團(tuán)隊(duì)在期刊《Bioactive Materials》上發(fā)表了題為“Engineered neutrophil apoptotic bodies ameliorate myocardial infarction by promoting macrophage efferocytosis and inflammation resolution”的文章,構(gòu)建了工程化中性粒細(xì)胞凋亡小體(eNABs)來模擬自然中性粒細(xì)胞凋亡,調(diào)節(jié)炎癥反應(yīng)并增強(qiáng)MI修復(fù)。eNABs是通過結(jié)合具有良好炎癥傾向性和免疫調(diào)節(jié)特性的自然中性粒細(xì)胞凋亡體膜和負(fù)載有5-氨基乙酰丙酸己酯鹽酸鹽(HAL)的介孔二氧化硅納米顆粒制備的。eNABs主動(dòng)靶向巨噬細(xì)胞和組裝的HAL的同時(shí)啟動(dòng)血紅素生物合成途徑,在細(xì)胞內(nèi)釋放后產(chǎn)生抗炎膽紅素,從而進(jìn)一步增強(qiáng)抗炎作用。在體內(nèi)研究中,eNABs有效調(diào)節(jié)梗死區(qū)域的炎癥反應(yīng)以改善心功能。這項(xiàng)研究證明了一種有效的仿生構(gòu)建策略來調(diào)節(jié)巨噬細(xì)胞的功能以進(jìn)行MI修復(fù)。
本文中使用了欣博盛大鼠TNF-α, IL-6, TGF-β and IL-10 ELISA試劑盒,檢測了大鼠巨噬細(xì)胞培養(yǎng)上清中細(xì)胞因子的含量。
