近期,溫州醫(yī)科大學(xué)的沈建良課題組、蔡曉軍課題組與鄧輝課題組合作在《Advanced Science》(IF=15.1)上發(fā)表題為“Cascade Reactions Catalyzed by Gold Hybrid Nanoparticles Generate CO Gas Against Periodontitis in Diabetes.”(金混合納米粒子催化級聯(lián)反應(yīng)產(chǎn)生一氧化碳氣體對抗糖尿病牙周炎)的研究,為未來治療糖尿病牙周炎提供了有益的啟示。
由于糖尿病牙周炎存在局部炎癥,其特點是葡萄糖濃度過高、細菌感染和高氧化應(yīng)激,因此治療糖尿病牙周炎是一項重大挑戰(zhàn)。在該項研究中,研究團隊將介孔二氧化硅納米顆粒(MSN)綴合了金納米顆粒(Au NPs)并負載了羰基錳,從而制備出一種一氧化碳(CO)增強型多酶協(xié)同雜化納米平臺(MSN-Au@CO)。Au NPs的類葡萄糖氧化酶活性可催化葡萄糖氧化成過氧化氫(H2O2)和葡萄糖酸,然后通過類過氧化物酶活性將 H2O2 轉(zhuǎn)化為羥自由基(-OH),從而消滅細菌。此外,H2O2產(chǎn)生的CO與Au NPs對受到脂多糖攻擊的巨噬細胞具有協(xié)同抗炎作用。其根本機制可能是誘導(dǎo)核因子紅細胞2相關(guān)因子2(nuclear factor erythroid 2-related factor 2)減少活性氧,并抑制核因子kappa-B信號以減輕炎癥反應(yīng)。重要的是,MSN-Au@CO的抗菌和抗炎作用在患有結(jié)扎誘發(fā)的牙周炎的糖尿病大鼠模型中(ligature-induced periodontitis)得到了驗證,證實了牙周骨質(zhì)流失減少且具有良好的生物相容性??傊?,MSN-Au@CO利用葡萄糖激活級聯(lián)反應(yīng)消滅細菌,并與氣體療法(gas therapy)協(xié)同調(diào)節(jié)免疫微環(huán)境,為糖尿病牙周炎的治療提供了一個潛在的方向。
