肥胖癥是一種慢性代謝性疾病,肥胖可導致一系列嚴重的并發(fā)癥,比如冠心病、糖尿病、高血壓、脂肪肝、癌癥等,目前肥胖癥的主要治療方法包括生活方式干預、藥物治療和手術治療,治療機制多種多樣,但是每種方法有自己的缺點,并且由于多種因素,減肥效果有限。因此,迫切需要尋找安全有效創(chuàng)新的方法來對抗肥胖。光動力療法和脂肪棕化誘導是兩種獨特的減肥方法,前者起效迅速但范圍小,后者作用相對緩慢但影響廣泛。如果將這兩種策略結(jié)合起來治療肥胖癥將有重要意義。2021年2月,廈門大學聶立銘教授團隊在國際知名期刊Nature Nanotechnology(影響因子:31.538)上在線發(fā)表了題目為“Photoacoustic molecular imaging-escorted adipose photodynamic-browning synergy for fighting obesity with virus-like complexes”的文章,提出了一種新穎的光聲分子成像護航下的光動力/棕化抗肥胖策略,為肥胖治療及療效評估提供了一個新思路。
本研究通過基因工程技術改造了乙肝病毒核心蛋白顆粒,將可追蹤的光敏劑酞菁鋅四磺酸鋅ZnPcS4和褐變劑羅格列酮(rosiglitazone)結(jié)合到乙型肝炎脂肪靶向蛋白復合結(jié)構中,于肥胖小鼠中靜脈注射后,該復合物特異性結(jié)合白色脂肪組織,尤其是那些血液供應豐富的組織,并由于ZnPcS4光動力學和羅格列酮褐變的協(xié)同作用而促使脂肪減少。使用光聲分子成像,能夠可視化監(jiān)測脂肪可能發(fā)生的效應變化。
本研究中使用了欣博盛TNF-α, IL-1β and IL-6 ELISA試劑盒檢測了小鼠白色脂肪組織勻漿中細胞因子的濃度。
