近期，中國科學(xué)院上海藥物研究所于海軍老師課題組在國際權(quán)威學(xué)術(shù)期刊Advanced materials上發(fā)表了題為“Engineering Stimuli-Activatable Boolean Logic Prodrug Nanoparticles for Combination Cancer Immunotherapy”的論文，本文首次報(bào)道了一種用于免疫調(diào)節(jié)劑（如免疫激活劑和免疫抑制劑）的腫瘤靶向協(xié)同輸送及聯(lián)合免疫治療的布爾邏輯前藥納米粒（BLPNs）。構(gòu)建了一個(gè)刺激激活BLPNs庫，通過調(diào)整輸入組合包括腫瘤微環(huán)境中的細(xì)胞外基質(zhì)金屬蛋白2/9（MMP-2/9）、細(xì)胞內(nèi)酸度（pH=5.0-6.0）和還原型谷胱甘肽，來產(chǎn)生YES/AND邏輯輸出。通過指定正交刺激-不穩(wěn)定部分的分子結(jié)構(gòu)和連接性，同時(shí)利用腫瘤部位內(nèi)源性信號(hào)，實(shí)現(xiàn)對(duì)BLPNs分解和前藥活化的選擇性可調(diào)控制，進(jìn)一步證明了BLPNs在腫瘤中的特異性分布和高效腫瘤免疫治療中免疫調(diào)節(jié)劑的刺激激活作用。本研究結(jié)果為腫瘤藥物特異性輸送和精準(zhǔn)的癌癥免疫治療開辟了新的途徑。
本研究使用欣博盛IL-6, TNF-ɑ和IFN-γ ELISA試劑盒檢測了小鼠腫瘤組織樣本中的細(xì)胞因子的含量。
