疫苗誘導(dǎo)的抗腫瘤免疫應(yīng)答速率過慢無法有效抑制惡性腫瘤的快速生長(zhǎng),常常限制了腫瘤治療性疫苗的效率,如何快速誘導(dǎo)并維持強(qiáng)烈的抗腫瘤免疫應(yīng)答是當(dāng)前腫瘤治療性疫苗研究待解決的難點(diǎn)之一。許多功能化的微/納米疫苗被證明能加強(qiáng)腫瘤免疫治療,但很少有研究將這些微/納米疫苗的治療效果與疫苗接種途徑(如靜脈注射、皮下注射或兩者的組合)聯(lián)系起來。近日,中國(guó)科學(xué)院過程工程研究所劉瑞田研究員、昆士蘭大學(xué)澳大利亞生物工程與納米技術(shù)研究所許志平教授和張凌宵博士等人在《Nano Today》(IF=16.907)上發(fā)表了題為”Nanovaccine’s rapid induction of anti-tumor immunity significantly improves malignant cancer immunotherapy”的研究論文,首次提出聯(lián)合靜脈免疫(IV-VAC)和皮下免疫(SC-VAC)的腫瘤疫苗接種新策略,成功實(shí)現(xiàn)強(qiáng)烈的抗腫瘤免疫應(yīng)答的快速誘導(dǎo)與維持。本文采用經(jīng)典的“啟動(dòng)+促進(jìn)”免疫策略,研究了4種納米疫苗組合(IV+IV、IV+SC、SC+IV和SC+SC)對(duì)E.G7-OVA淋巴瘤和B16F10黑色素瘤小鼠抗腫瘤治療效果的影響。通過將抗原和CpG加載到層狀雙金屬氫氧化物(Layered Double Hydroxide, LDH)納米粒子上構(gòu)建納米疫苗。實(shí)驗(yàn)數(shù)據(jù)表明,采用“IV初免 + SC加強(qiáng)”(IV + SC)的疫苗接種策略(使用IV-VAC進(jìn)行初次免疫,隨后使用SC-VAC進(jìn)行免疫加強(qiáng)),能夠在腫瘤早期快速誘導(dǎo)機(jī)體產(chǎn)生強(qiáng)烈的抗腫瘤免疫應(yīng)答來抑制腫瘤生長(zhǎng),并在SC-VAC的幫助下逐步加強(qiáng)免疫強(qiáng)度。


本文中使用了欣博盛Mouse IFN-γ and IL-4 ELISA試劑盒檢測(cè)了小鼠脾細(xì)胞分泌細(xì)胞因子的水平。
