細(xì)胞外囊泡(EVs)在疾病治療和藥物遞送方面具有廣闊的應(yīng)用潛力。然而,如何實現(xiàn)準(zhǔn)確從EVs釋放藥物以及EVs與藥物在靶組織中的協(xié)同治療效果仍具有很大的挑戰(zhàn)性。近日,北京理工大學(xué)謝海燕教授課題組在期刊Advanced Materials上發(fā)表了題為“Self-Activatable Photo-Extracellular Vesicle for Synergistic Trimodal Anticancer Therapy.”的文章,報道了一種工程化自激活的光-EVs,并將其用于協(xié)同三模態(tài)抗癌治療。
M1巨噬細(xì)胞來源的EVs (M1 EVs)可同時負(fù)載雙[2,4,5-三氯-6-(戊氧羰基)苯基]草酸酯(CPPO)、Ce6和前藥Dox-EMCH。給藥后,由于M1 EVs具有腫瘤歸巢能力,因此該M1 EVs系統(tǒng)能夠主動靶向腫瘤細(xì)胞,并且可將M2重極化為M1巨噬細(xì)胞,因此其不僅表現(xiàn)出了免疫治療作用,還能夠產(chǎn)生H2O2。H2O2和CPPO之間的反應(yīng)能夠產(chǎn)生激活Ce6的化學(xué)能,隨后既可以產(chǎn)生用于成像的化學(xué)發(fā)光,也能夠產(chǎn)生用于光動力治療(PDT)的單線態(tài)氧(1O2)。同時,1O2誘導(dǎo)膜破裂可以實現(xiàn)Dox-EMCH的釋放,其在被激活后可以穿透至腫瘤深部的乏氧區(qū)。實驗結(jié)果表明,免疫治療、PDT和化療產(chǎn)生的協(xié)同作用可以產(chǎn)生很好的抗癌效果,在對抗癌癥方面具有不錯的應(yīng)用前景。

本文中使用了欣博盛小鼠TNF-α and IL-6 ELISA Kits檢測了巨噬細(xì)胞分泌細(xì)胞因子的水平。
