免疫穩(wěn)態(tài)失衡可以導(dǎo)致自身免疫疾病的發(fā)生。調(diào)節(jié)性T細(xì)胞（Tregs）是一種具有誘導(dǎo)和維持免疫耐受功能的T細(xì)胞亞群，在預(yù)防自身免疫疾病和維持免疫穩(wěn)態(tài)中起核心作用，可用于免疫穩(wěn)態(tài)失衡的治療。然而，在體內(nèi)使用非特異性Tregs治療自身免疫疾病可能增加患者全身免疫抑制的風(fēng)險(xiǎn)，因此，誘導(dǎo)抗原特異性Tregs是疾病治療的最佳選擇。在利用Tregs建立新的免疫穩(wěn)態(tài)之前，首先應(yīng)清除過多的活化T細(xì)胞，打破已失衡的免疫狀態(tài)。以往研究通過輻射或使用抗體來破壞失衡的免疫響應(yīng)，但此類方法存在很大的脫靶風(fēng)險(xiǎn)，并可能導(dǎo)致免疫缺陷。此外，抗原特異性免疫耐受的有效誘導(dǎo)需要T細(xì)胞編程的精確協(xié)調(diào)，這是很難實(shí)現(xiàn)的。
2021年3月11日，西安交通大學(xué)陳鑫教授團(tuán)隊(duì)與空軍軍醫(yī)大學(xué)金巖、劉世宇教授團(tuán)隊(duì)合作在Science Translational Medicine以封面形式發(fā)表了題為Modular immune-homeostatic microparticles promote immune tolerance in mouse autoimmune models 的文章。該研究成功構(gòu)建了一種通過清除活化T細(xì)胞誘導(dǎo)免疫耐受的多級(jí)微納材料，并基于模塊化設(shè)計(jì)，在不同自身免疫疾病模型中誘導(dǎo)抗原特異性Tregs，為自身免疫疾病的治療提供了新策略。
本文中使用了欣博盛ELISA試劑盒分別檢測(cè)了小鼠血清中TGF-β1, IL-17, IL-6和TNF-α等細(xì)胞因子的濃度以及小鼠脾細(xì)胞培養(yǎng)上清中IL-17, IL-6, TNF-α 和IFN-γ的濃度。
