先天免疫是宿主抵御病毒感染的第一道防線。 病毒成分(尤其是其基因組或復(fù)制中間體)會(huì)被不同的模式識(shí)別受體(PRRs)感知,包括Toll樣受體(TLRs)、RIG-I樣受體(RLRs)和/或細(xì)胞內(nèi)DNA傳感器。PRRs對(duì)病毒核酸的感知會(huì)觸發(fā)信號(hào)級(jí)聯(lián),從而誘導(dǎo)I型干擾素(IFN)、促炎細(xì)胞因子和其他下游抗病毒效應(yīng)基因的轉(zhuǎn)錄。下游的細(xì)胞因子和效應(yīng)物抑制病毒復(fù)制或誘導(dǎo)受感染細(xì)胞的凋亡,導(dǎo)致先天性抗病毒反應(yīng)。但是體液代謝是否以及如何調(diào)節(jié)先天抗病毒反應(yīng)仍然是個(gè)謎。最近,武漢大學(xué)中南醫(yī)院醫(yī)學(xué)研究所免疫與代謝前沿科學(xué)中心在期刊《Signal Transduction and Targeted Therapy》(IF=18.187)上發(fā)表題為“Modulation of innate immune response to viruses including SARS-CoV-2 by progesterone.”的文章,發(fā)現(xiàn)病毒感染通過小鼠下丘腦-垂體-腎上腺軸誘導(dǎo)孕酮。孕酮誘導(dǎo)下游抗病毒基因并促進(jìn)細(xì)胞和小鼠的先天抗病毒反應(yīng),而敲除孕酮受體PGR具有相反的效果。孕酮刺激PGR激活酪氨酸激酶SRC,使轉(zhuǎn)錄因子IRF3在Y107磷酸化,導(dǎo)致其激活并誘導(dǎo)抗病毒基因。感染新型冠狀病毒病毒的患者孕酮水平升高,這與新冠肺炎的嚴(yán)重程度降低相關(guān)。該發(fā)現(xiàn)揭示了黃體酮如何調(diào)節(jié)宿主先天抗病毒反應(yīng),并指出黃體酮是一種潛在的感染性和炎性疾病的免疫調(diào)節(jié)劑。
