急性肺損傷(ALI)包括肺部對(duì)炎癥或化學(xué)損傷的統(tǒng)一反應(yīng)，通常由全身性疾病引起，包括敗血癥或創(chuàng)傷、病原體感染和有毒氣體吸入。盡管有大量研究如何治療感染的免疫反應(yīng)，但由于新病原體的出現(xiàn)(如新型冠狀病毒肺炎的全球疫情)以及耐藥性的持續(xù)上升，仍然沒(méi)有有效的藥物療法來(lái)治療。內(nèi)皮細(xì)胞激活在膿毒癥誘導(dǎo)的急性肺損傷的發(fā)病機(jī)制中起著重要作用，然而，詳細(xì)的調(diào)節(jié)機(jī)制仍不清楚。最近，南昌大學(xué)轉(zhuǎn)化醫(yī)學(xué)研究所在期刊《Signal Transduction and Targeted Therapy》（IF=18.187）上發(fā)表題為“TRIM47 is a novel endothelial activation factor that aggravates lipopolysaccharide-induced acute lung injury in mice via K63-linked ubiquitination of TRAF2.”的文章，報(bào)道了TRIM47，一種含有三部分基序的蛋白家族的E3泛素連接酶，在血管內(nèi)皮細(xì)胞中高度表達(dá)。TRIM47缺陷小鼠通過(guò)減輕肺部炎癥能有效抵抗脂多糖(LPS)誘導(dǎo)的急性肺損傷和死亡。TRIM47在體外TNFα誘導(dǎo)的內(nèi)皮細(xì)胞活化過(guò)程中上調(diào)。在內(nèi)皮細(xì)胞中敲低TRIM47抑制多種促炎細(xì)胞因子的轉(zhuǎn)錄，減少單核細(xì)胞粘附和粘附分子的表達(dá)，并抑制內(nèi)皮細(xì)胞中IL-1β和IL-6的分泌。而TRIM47的過(guò)度表達(dá)促進(jìn)了TNFα刺激下的炎癥反應(yīng)和單核細(xì)胞粘附。研究表明，TRIM47作為一種新的內(nèi)皮細(xì)胞激活劑，通過(guò)增強(qiáng)TRAF2的K63相關(guān)泛素化，進(jìn)而激活NF-κB和MAPK信號(hào)通路，觸發(fā)內(nèi)皮細(xì)胞的炎癥反應(yīng)，從而促進(jìn)LPS誘導(dǎo)的肺部炎癥和急性肺損傷。
本文中使用了欣博盛Human TNFα, IL-1β and IL-6 ELISA kits檢測(cè)了血清和細(xì)胞培養(yǎng)液上清樣本中的表達(dá)量。
