暴露于生長因子豐富的環(huán)境對器官疾病、組織損傷的再生，例如骨骼系統(tǒng)缺陷和骨再生具有顯著的恢復(fù)作用。盡管已經(jīng)有研究報道了將某些生長因子引入相關(guān)領(lǐng)域，但是它們的再生效果不令人滿意。最近，華東理工大學(xué)在期刊《Bioactive Materials》（IF=16.874）上發(fā)表題為 “Recapitulation of growth factor-enriched microenvironment via BMP receptor activating hydrogel.” 的文章，研究出一個生長因子富集的微環(huán)境(GEM)的創(chuàng)新水凝膠。這種硫酸化水凝膠包含了骨形態(tài)發(fā)生蛋白-2 (BMP-2)，這是成骨過程中的一種重要生長因子，可指導(dǎo)間充質(zhì)干細胞(MSC)分化，刺激細胞增殖，并改善骨形成。半合成水凝膠磺化明膠(S-Gelatin)可以通過增強BMP-2和BMP-2 II型受體(BMPR2)之間的結(jié)合來放大小鼠MSCs中的BMP-2信號，這些受體位于MSC細胞核上并被水凝膠激活。研究結(jié)果顯示MSCs在整個再生過程中細胞因子分泌的顯著改善證實了S-Gelatin/rhBMP-2水凝膠獲得生長因子的潛力，促進血管形成。這個研究成果提供了一種新的策略，通過放大rhBMP-2的再生能力和捕獲內(nèi)源性生長因子來實現(xiàn)原位GEM和加速骨再生。
本文中使用了欣博盛Human BMP-2 ELISA kit檢測了材料中釋放的BMP-2的表達量。
