在藥物開發(fā)的早期階段,尋找novel scaffolds化合物是一項(xiàng)重要但具有挑戰(zhàn)性的任務(wù)。目前已經(jīng)提出了各種模型來產(chǎn)生藥物樣分子,然而這些模型設(shè)計(jì)具有novel scaffolds化合物的wet-lab驗(yàn)證和靶特異性分子的能力幾乎沒有得到驗(yàn)證。最近,四川大學(xué)華西醫(yī)院的研究團(tuán)隊(duì)在期刊《Nature Communications》(IF=17.694)上發(fā)表題為“Generative deep learning enables the discovery of a potent and selective RIPK1 inhibitor.”的文章,提出了一種生成式深度學(xué)習(xí)(GDL)模型,一種分布學(xué)習(xí)條件遞歸神經(jīng)網(wǎng)絡(luò)(cRNN),用于為給定的生物靶標(biāo)生成定制的虛擬化合物庫。然后將GDL模型應(yīng)用于RIPK1。對(duì)生成的特制化合物庫進(jìn)行虛擬篩選和隨后的生物活性評(píng)估,發(fā)現(xiàn)了一種有效的選擇性RIPK1抑制劑,具有以前未報(bào)道的scaffolds RI-962。該化合物在保護(hù)細(xì)胞免于壞死方面顯示出有效的體外活性,并且在兩種炎癥模型中顯示出良好的體內(nèi)功效。這些發(fā)現(xiàn)證明了研究工作者的GDL模型在利用未報(bào)道的scaffolds生成hit/lead化合物方面的能力,突出了深度學(xué)習(xí)在藥物發(fā)現(xiàn)中的巨大潛力。

本文中使用了欣博盛Mouse TNFα, IL-6, and IL-1β ELISA kits檢測(cè)了結(jié)腸組織勻漿樣本中這些細(xì)胞因子的含量。
