合成生物學的快速發(fā)展激發(fā)了人們對能夠診斷和治療疾病的工程微生物的興趣。然而,設計檢測動態(tài)疾病相關生物標志物的細菌,然后進行驅(qū)動治療仍然很困難。最近,華東理工大學的研究團隊在期刊《Cell Host & Microbe》(IF=31.316)上發(fā)表題為“Biomarker-responsive engineered probiotic diagnoses, records, and ameliorates inflammatory bowel disease in mice.”的文章,他們開發(fā)了一種工程益生菌,它可以非侵入性地實時監(jiān)測和記錄炎癥性腸病(inflammatory bowel disease,IBD)的發(fā)生和發(fā)展,并可以通過一種自我調(diào)節(jié)的機制來響應這些生物標志物,從而釋放治療藥物。這些用于診斷和治療的智能響應細菌(i-ROBOT)由大腸桿菌Nissle 1917組成,該細菌通過激活堿基編輯系統(tǒng)來產(chǎn)生可遺傳的基因組DNA序列并產(chǎn)生比色信號,從而對炎癥標記物硫代硫酸鹽的水平做出響應。硫代硫酸鹽的波動也驅(qū)動免疫調(diào)節(jié)劑AvCystatin的可調(diào)釋放。給患有結(jié)腸炎的小鼠口服i-ROBOT,在處理過的糞便和結(jié)腸樣品中產(chǎn)生分子記錄信號,并有效改善疾病。i-ROBOT為胃腸和其他代謝疾病提供了一個有前景的設計思路。
本文中使用了欣博盛Human IL-10和IL-12/23 p40 ELISA kits檢測了細胞培養(yǎng)液上清樣本中這些細胞因子的含量。
