對(duì)乙酰氨基酚(APAP)過量是藥物性肝損傷(DILI)的主要原因。但是目前腸道微生物群和相關(guān)代謝物對(duì)APAP和肝功能的影響仍不清楚。最近,南方醫(yī)科大學(xué)團(tuán)隊(duì)在期刊《Cell Host & Microbe》(IF=31.316)上發(fā)表題為“Liberation of daidzein by gut microbial β-galactosidase suppresses acetaminophen-induced hepatotoxicity in mice.”的文章,該篇研究發(fā)現(xiàn)APAP紊亂與獨(dú)特的腸道微生物群落有關(guān),即陰道乳酸桿菌明顯減少。由于細(xì)菌β-半乳糖苷酶從飲食中釋放異黃酮大豆苷元,接受陰道毛滴蟲的小鼠顯示出對(duì)APAP肝毒性的抗性。用β-半乳糖苷酶抑制劑消除了陰道毛滴蟲對(duì)APAP暴露的無菌小鼠的肝保護(hù)作用。類似地,β-半乳糖苷酶缺陷型陰道毛滴蟲在APAP處理的小鼠中比野生型小鼠產(chǎn)生更差的結(jié)果,但是這些差異通過大豆苷元給藥得以克服。大豆黃酮阻止了鐵死亡,這與法尼二磷酸合酶(Fdps)表達(dá)下降有關(guān),該合酶激活了涉及AKT-GSK3β-Nrf2的關(guān)鍵鐵死亡途徑。該研究證明了陰道乳酸桿菌β-半乳糖苷酶釋放大豆苷元抑制了Fdps介導(dǎo)的肝細(xì)胞鐵死亡,為藥物性肝損傷提供了新的治療方法。
本文中使用了欣博盛Mouse TNF-α, MCP-1 和IL-6 ELISA kits檢測了血漿中這些細(xì)胞因子的含量。
