肺轉移是癌細胞從我們身體其他地方發(fā)展的原發(fā)性腫瘤擴散到肺，這被認為是癌癥死亡的關鍵原因。其治療受到轉移性腫瘤中有限的藥物遞送效率和免疫抑制的限制。最近，中國科學院上海藥物研究所的研究團隊在期刊《ACS nano》（IF=18.027）上發(fā)表題為 “Spatial-Drug-Laden Protease-Activatable M1 Macrophage System Targets Lung Metastasis and Potentiates Antitumor Immunity.” 的文章，該研究設計了一個空間藥物加載的M1巨噬細胞系統(tǒng)，其內部是脂質體R848，在M1巨噬細胞(RDM)的膜上是成纖維細胞活化蛋白蛋白酶(FAP)敏感的磷脂-DM4綴合物。RDM可優(yōu)先積聚在肺轉移病灶處，并作為游離藥物分子或載藥納米囊泡響應性地釋放治療劑。與陰性對照組相比，RDM治療顯著增強了CD3+ CD8+ T細胞對肺轉移的浸潤，并分別導致顆粒酶-B-、干擾素-γ和Ki67-陽性亞型增加8.54倍、12.87倍和2.85倍。此外，RDM治療在4T1模型中產生了90.99%的肺轉移抑制，并在三個鼠肺轉移模型中顯著延長了生存期。該載藥體系為靶向肺轉移和增強抗腫瘤免疫用于抗轉移治療提供了可行策略。
本文中使用了欣博盛Mouse TNF-α, IL-1β 和IL-6 ELISA kits檢測了細胞培養(yǎng)上清中這些細胞因子的含量。
