經(jīng)典的(caspase-1)和非經(jīng)典的(包括caspase-4、5和11)在炎癥小體中均切割gasdermin D (GSDMD)誘導(dǎo)細(xì)胞焦亡。caspase-1加工處理IL-1β和IL-18使其成熟 , 而目前還沒有確定脂多糖激活的caspase-4、5和11細(xì)胞因子靶點(diǎn)。最近,北京生命科學(xué)研究所研究團(tuán)隊(duì)在期刊《Nature》(IF=64.8)上發(fā)表題為“Recognition and maturation of IL-18 by caspase-4 noncanonical inflammasome.”的文章,該研究團(tuán)隊(duì)的研究結(jié)果表明顯示激活的人caspase-4,而不是小鼠 caspase-11在體外和細(xì)菌感染過程中直接加工IL-18 。caspase-4切割pro-IL-18中與caspase-1相同的四肽位點(diǎn)。 caspase-4-pro-IL-18復(fù)合物的晶體結(jié)構(gòu)揭示了一個(gè)雙位點(diǎn)(二元) 底物識(shí)別機(jī)制。這種二元識(shí)別也被caspase-5和caspase-1用來(lái)加工pro-IL-18。這項(xiàng)研究發(fā)現(xiàn)并證明炎癥因子IL-18是非經(jīng)典炎癥小體通路活化的caspase-4/5的底物,并完整地揭示了天然免疫通路中caspases識(shí)別和切割pro-IL-18,以及pro-IL-18被加工后成熟為有生理功能的炎癥因子的精確分子機(jī)制,為理解一些自身免疫疾病的發(fā)生機(jī)制以及開發(fā)新的治療方法提供了全新的思路。
本文中使用了欣博盛Human IL-18和 IFN-γ ELISA kits 檢測(cè)了細(xì)胞培養(yǎng)液上清中目標(biāo)蛋白的含量。
