近期，中國人民解放軍軍事醫(yī)學(xué)科學(xué)院李慧艷研究員團隊、張宇程副研究員團隊以及周濤研究員團隊合作在Cell（IF=64.5）上發(fā)表題為“Innervation of nociceptor neurons in the spleen promotes germinal center responses and humoral immunity”（脾臟痛覺神經(jīng)元的神經(jīng)支配促進生發(fā)中心反應(yīng)和體液免疫）的研究論文。在該項研究中，研究團隊證明了痛覺感受神經(jīng)纖維（nociceptive sensory nerve fibers）不僅沿著脾臟血管廣泛分布，而且能夠深入到B細(xì)胞區(qū)。脾神經(jīng)支配的痛覺感受器（nociceptors）主要來源于左側(cè)T8-T13背根神經(jīng)節(jié)（dorsal root ganglia, DRGs），促進脾生發(fā)中心（germinal center, GC）反應(yīng)和體液免疫。痛覺感受器可通過抗原誘導(dǎo)的脾前列腺素E2 （PGE2）積聚而被激活，釋放降鈣素基因相關(guān)肽（CGRP），進一步促進早期脾生發(fā)中心反應(yīng)。在機制上，CGRP通過其受體CALCRL-RAMP1經(jīng)環(huán)化AMP （cAMP）信號通路直接作用于B細(xì)胞。通過攝取辣椒素激活痛覺感受器可增強脾GC反應(yīng)和抗流感免疫。研究團隊建立了一種促進體液免疫的特異性DRG-脾臟感覺神經(jīng)連接，這表明通過靶向痛覺神經(jīng)系統(tǒng)來改善宿主免疫是可行的方法。
該項研究中，使用了欣博盛生物（NeoBioscience Technology Co, Ltd）的Mouse TNF-α ELISA試劑盒與Mouse IL-1β ELISA試劑盒，用于測定脾臟組織勻漿中相關(guān)細(xì)胞因子的含量。
